Hematoxylin is a brownish crystalline compound extracted from the heartwood of logwood. It is used as an indicator with a pH range of 5–6 and in the manufacture of ink, but is mainly used in oxidized form (called haematein) as a stain in microscopy. Hematoxylin is not itself the dye but conjuncted with a mordant, such as alum (ammonium, potassium, chrome. or ferric), ferric chloride, and phosphotungstic acid. These dyes are widely used as a stain in biology and medicine to demonstrate nuclear and cytoplasmic structures. These are also used to define and examine muscle fibers or connective tissue. The supply of hematoxylin is limited due to felling of forests. Some dyes are recommended as replacements such as Celestine blue (RN: 1562-90-9), Gallocyanine (CAS RN: 1562-85-2), Gallein (CAS RN: 2103-64-2), Solochrome cyanine R (CAS RN: 3564-18-9). But none of them are exactly recommended, Brazilin (Natural red 24) extracted from the wood of the brazilwood and and sappanwood is very close to hematoxylin. The structure differ is having one less hydroxyl group. Brazilin has been also used as a dye for fabric, and to make inks. Brazilein is the oxidized form of Brazilin.
