GENERAL DESCRIPTION & APPLICATIONS

Lipstatin, in IUPAC [(2S,4E,7E)-1-(3-hexyl-4-oxooxetan-2-yl)trideca-4,7-dien-2-yl] (2S)-2-formamido-3-methylpentanoate,is a irreversible inhibitor of pancreatic lipase, an enzyme that breaks down triglycerides in the intestine. Triglycerides are components of very low density lipoprotein (VLDL) and chylomicrons, which play an important role in metabolism as energy sources and transporters of dietary fat. Without pancreatic lipase, triglycerides are prevented from being hydrolyzed into absorbable free fatty acids and are excreted undigested. The saturated derivative of lipstatin ( tetrahydrolipstatin called orlistat) is the stable form enough to develop into a drug which reduce caloric intake by preventing the digestion, and therefore absorption of fats from the diet. Orlistat is used as an anti-obesity drug which decreases calorie intake and increases energy expanditure resulting in weight loss or control. Other types of anti-obesity drugs have suppression of the appetite. Catecholamines acting as stimulants are the main compounds used for this. So, anti-obesity drugs carry severe side effects including, life-threatening sometimes. Anti-obesity drugs are recommended only for short-term use under medical prescription.
